Early in his career, he worked at a variety of hospitals in New York, including Maimonides Hospital, Harlem Hospital, and Roosevelt Hospital. In 1969, he returned to Denver, Colorado, as a staff member of General Rose & Mercy Hospital. He continued his career at the Porter & Swedish College until 1993, becoming a director and chief anesthesiologist in 1980. The highlight of Dr. Clarke’s career was during a meeting for the state representative in Biloxi, Mississippi. One evening, a dinner was held for the National Conference of Black Legislatures. At his table, a young lady told him that she was going to lose her position because she was African American and the company she was working for was putting a lot of pressure on her. She was wondering if he would help her. He wrote letters to a segregationist senator and another representative; the girl who he vowed to help ultimately received her job back. Dr. Clarke was very happy to help her.

After retiring from medicine and before he went into the legislature, Dr. Clarke started minority-based chess clubs in elementary schools. During the 1960s in Englewood, New Jersey, is when he became involved in desegregation in elementary schools. In recognition of his career and work, he was featured in the Denver Post and on the Tom Brokaw show. He received the Best Politician award in 1997 and has received an additional accolade from the Westword News.
